以模式植物拟南芥(Columbia ecotype)为供试材料,利用脱色苯胺蓝染色的方法观察了拟南芥花粉管在雌蕊组织中的生长情况,获得了花粉管从萌发到进入花柱和子房不同时期的生长速率.结果表明,花粉管在不同时期的生长速率呈现出逐步加快的趋势.例时采用手工分离的方法分离了不同发育时期的胚胎,研究了胚胎发育随时间进程的形态结构特征,并与Landsberg生态型拟南芥的胚胎发育速率进行了比较,发现Columbia生态型的发育速率略慢.此外还测量了胚胎不同发育时期的角果长度,获得了胚胎发育时期与角果长度之间的对应关系,发现随着胚胎的发育,角果的生长速率晕现出逐渐递减的趋势.
With the aid of aniline blue staining, pollen tube growth in pistil tissue was observed in Arabidopsis thaliana (Columbia ecotype), and the growth speed of pollen tubes in style and ovary were calculated, the results showed that the pollen tube grew faster as time goes on. Furthermore, embryos at different developing stages were manually isolated to understand their changes of morphologic character along with the time course, then the developmental situation difference were compared between the Columbia ecotype and the Landsberg ecotype, the results showed that the developmental speed of the Columbia ecotype was slower than that of the Landsberg ecotype. Besides, the lengths of siliques at different developing stages were measured, and relation between silique length and time course were investigated, the results showed that the siliques grew slower as time goes on. The results could be valuable and reliable information for the reproductive developmental biology researches of the model plant Arabidopsis thaliana.
