目的:通过观察电针对食源性肥胖大鼠中枢及胰腺组织中磷酸化腺苷酸激活蛋白激酶仪(pAMPKα)表达的影响,探讨针灸减肥的可能机制。方法:sD大鼠50只,随机分成正常组8只和高脂组42只,造模12周后挑选体重超过正常组体重15%的大鼠16只,随机分为电针治疗组(电针组)8只及肥胖模型组(模型组)8只,电针组大鼠针刺足三里、内庭穴位减肥(2/15Hz,3mA),每次15min,每天1次,共30次。治疗结束后,采用免疫组织化学方法测定各组下丘脑腹内侧核(VMN)及胰腺组织pAMPKα的表达。结果:电针组与模型组比较,体重明显下降(P〈0.01),与正常组体重相近(P〉0.05)。电针组大鼠胰腺中pAMPKd平均光密度明显高于模型组泸〈0.01),与正常组相近p〉0.05);各组下丘脑腹内侧核中pAMPKd平均光密度差异无显著性意义(P〉0.05)。结论:电针可通过升高胰腺pAMPKα,即增加能量消耗达到治疗食源性肥胖的目的。
Objective: To observe the effect of electro-acupuncture(EA) on expression of central and pancreatic phosphory- lation AMPKα (pAMPKα) in diet-induced obesity(DIO) rats and to explore the possible mechanism of acupuncture in reducing weight. Methods. Fifty SD rats were randomly divided into the normal group(n=8) and DIO model group(n=42).After modeling for 12 weeks, 16 rats with overweight by 15% of the normal rats were randomly divided into EA group(n=8) and the model group (n=8). EA group rats were treated by EA(2/15Hz, 3mA) at Zusanli (ST36) and Neiting(ST45) for 15 minutes each time, once a day, 30 times in total. After treatment, the expression of pAMPKα in ventromedial hypothalamic nucleus(VMN) and pancreatic tissue in each group was detected by immunohistochemical method. Results: Body weight of EA group was remarkably decreased(P 〈 0.01 compared with that of the model group), and was similar to that of the normal group(P 〉 0.05). The average optical density of pancreatic pAMPKα in EA group was significantly, higher than that in the model group(P 〈 0,01), while was similar to that in normal group(P 〉 0.05). For the average optical density of pAMPKα in VMN, there was no significant difference among three groups(P 〉 0.05). Conclusion. The therapeutic mechanism of EA in treating DIO is probably related with the increase of energy expenditure through increasing pancreatic pAMPKα.
