目的探讨Vps4A对肝癌细胞SMMC-7721中Wnt途径的影响。方法构建过表达Vps4A的SMMC-7721细胞稳定株,通过RT-q PCR的方法检测19个WNT基因的转录水平的变化,进一步通过Western blot及检测相关下游基因Notch2的变化进行确定。结果在SMMC-7721中,经典Wnt亚型Wnt10a和Wnt10b的m RNA水平较高,而在过表达Vps4A之后,其蛋白及m RNA水平均显著下降。同时,在过表达Vps4A细胞株中,与经典Wnt通路相关的下游靶基因Notch2在转录及表达水平同样明显下调。结论Vps4A选择性抑制肝癌细胞SMMC-7721的经典Wnt/Notch2途径。
Objective To explore the effects of Vps4 A on Wnt signaling in hepatocellular carcinoma SMMC-7721 cells. Methods SMMC-7721 stable cell lines expressing Vps4 A was generated. Then,the m RNA levels of total 19 Wnt subtypes were tested using RT-q PCR. The results were further confirmed by Western blot and investigating the downstream gene Notch2. Results Wnt10 a and Wnt10 b were highly expressed in SMMC-7721 cells, and Vps4 A overexpressing significantly reduced their expression. Additionally, the levels of protein and m RNA of their downstream gene Notch2 were also obviously decreased. Conclusion Overexpressing Vps4 A inhibits the canonical Wnt / Notch2 signaling in SMMC-7721 cell.
