目的观察低浓度混合酶改良消化法体外培养SD大鼠面颌肌细胞的结果。方法分离SD大鼠颌面部咬肌区肌肉组织,先用0.1%的Ⅰ型胶原酶消化后再用0.1%的Ⅰ型胶原酶和0.125%的胰蛋白酶(1:1)低浓度混合酶消化法分离细胞,差速贴壁法纯化,所得细胞进行结蛋白(Desmin)免疫组化染色。结果体外培养所得细胞Desmin胞质阳性率达95%。结论采用低浓度混合酶消化和差速贴壁法体外培养SD大鼠面颌肌细胞可获得纯度高、产量高、活性强、稳定性好的面颌肌细胞。
Objective To observe the cuhure of maxillofacial skeletal muscle cells of SD rat's in vitro by method of improved low concentrations hybrid enzymatic dissociation. Methods The maxillofacial masseter muscle tissue was separa- ted from neonatal SD rat's, digested with 0.1% type Ⅰ collagenase and then with low concentration of 0.1% collagenase type Ⅰ and 0. 125% trypsin ( 1 : 1 ) hybrid enzyme to isolated cells, purified by differential adherence method, stained by immunohistochemical of desmin. Results The Desmin cytoplasmic positive rate was 95% in cultured cells. Conclusion The high purity, high yield, strong activity, stability, and good stability maxillofacial skeletal muscle myoblasts are easy to obtain from SD rat's in vitro by method of low concentration of mixed enzyme digestion and differential adhesion.
