目的 观察活血化瘀中药汤剂对糖尿病肾病(DN)大鼠模型肾间质纤维化的影响,探讨其可能的作用机制。方法 选用健康雄性SD大鼠30只,采用高糖高脂饲料喂养、一次性腹腔注射链脲佐菌素(STZ)诱导建立糖尿病SD大鼠模型,然后平分为DN模型组、活血化瘀组和厄贝沙坦组各10只,活血化瘀组选择活血化瘀中药汤剂灌胃服药,厄贝沙坦组选择厄贝沙坦溶液灌胃服药,模型组灌服等体积0.9%氯化钠注射液,记录三组肾间质纤维化相关指标的变化情况。结果 所有大鼠都造模成功,活血化瘀组与厄贝沙坦组大鼠的肾质量都明显低于DN模型组(P〈0.05)。实验后活血化瘀组与厄贝沙坦组组大鼠BUN、Scr较DN模型组明显降低(P〈0.05)。实验后DN模型组、活血化瘀组和厄贝沙坦组大鼠尿白蛋白排泄率(UAER)值分别为(16.32±4.29)mg/24 h、(11.50±2.59)mg/24 h和(12.20±3.01)mg/24 h,活血化瘀组和厄贝沙坦组较DN模型组明显降低(P〈0.05)。TGF-β1主要在肾间质区、肾小管部位及小动脉中表达;FN主要表达在肾小球、细胞外基质和肾小管部位。Western蛋白印迹检测显示活血化瘀组和厄贝沙坦组的FN和TGF-β1蛋白表达水平明显低于DN模型组大鼠(P〈0.05)。结论 活血化瘀中药汤剂能够降低DN大鼠的肾脏肥大,提高肾功能,改善肾组织病变,通过抑制肾组织TGF-βl与FN的表达,延缓肾间质纤维化。
Objective: To observe the effects of Huoxue Huayu decoction on kidney interstitial fibrosis in dia- betic nephropathy rats' model to explore its possible mechanism. Methods: From January 2015 to July 2015,30 clean grade healthy male Sprague Dawley rats were fed with high glucose and high fat diet and injected with streptozotocin (STZ) for inducing diabetic SD rats' model,and then were equally divided into DN model group, irbesartan group and Huoxue Huayu group with 10 rats in each group. The Huoxue Huayu group was given Huoxue Huayu decoction by intragastric administration,the irbesartan group was given irbesartan solution by intragastric administration,the DN model group was fed with normal saline of the same volume,and the changes on the renal interstitial fibrosis related indexes were recorded in the three groups. Results: All rats were successfully modeled. The rat kidney weight in the Huoxue Huayu group and irbesartan group were significantly lower than that of the DN model group (P〈0.05). After the experiment,the serum BUN and Set level in the Huoxue Huayu group and irbesartan group were significantly lower than that of the DN model group (P〈 0.05). After the experiment,the rats' UAER in the DN model group,Huoxue Huayu group and irbesartan group were (16.32±4.29)mg/ 24 h, (11.50±2.59) mg/24 h and (12.20 ±3.01 ) mg/24 h. The value of Huoxue Huayu group and irbesartan group showed significant difference with the DN model group(P〈0.05). The TGF-β1 was mainly expressed in the renal interstitial region, renal tubular part and small artery, and FN was mainly expressed in the glomerular, extracellular matrix and renal tubular parts. Western blot showed that the expression level of TGF-β1 and FN protein in Huoxue Huayu group and irbesartan group were significantly lower than that of DN rats in the model group (P〈 0.05).
