目的:本研究利用电刺激右侧三叉神经节区构建大鼠偏头痛模型,观察电针预处理对偏头痛大鼠内源性大麻素CB1受体、C-fos蛋白及m RNA表达,探讨电针预处理防治偏头痛作用的相关作用机制。方法:SD大鼠随机数字表法分为4组:假手术(A)组、模型(B)组、电针预处理+模型(C)组、利莫那班拮抗剂+电针预处理+模型(D)组。采用电刺激右侧三叉神经节制备偏头痛模型。以电针风池、外关预处理为被试因素,通过免疫组化法检测大鼠三叉神经尾核CB1受体和C-fos蛋白及m RNA表达。结果:与A组相比,B组、C组、D组CB1受体、C-fos蛋白及m RNA表达显著上调(P〈0.05);与B组相比,C组、D组CB1受体蛋白及m RNA表达显著上调(P〈0.05),C-fos蛋白及m RNA表达显著下调(P〈0.05);C组及D组间CB1受体蛋白及m RNA表达差异无统计学意义,而C-fos蛋白及m RNAC组较D组表达显著下调(P〈0.05)。结论:电针预处理可上调CB1受体蛋白及m RNA表达,下调C-fos蛋白及m RNA表达,提示电针预处理通过CB1受体减弱三叉神经初级传入纤维传入的兴奋信号,抑制三叉神经血管系统激活。这可能是电针预处理防治偏头痛作用机制之一。
Objective:To investigate the molecular biology basis on electroacupuncture pretreatment for migraine prophylaxis and to study the mechanism of electroacupuncture pre-disposal for migraine prophylaxis by assaying protein and mRNA expression of CB1 receptor and C-fos in the trigeminal nucleus caudalis in animal models of migraine.Methods:SpragueDawley rats were randomly divided into four groups:sham operation group,model group,electroacupuncture pretreatment,rimonabant antagonist group.The animal model of rats with migrane was established by right electrical stimulation of the trigeminal ganglion.The electroacupuncture pretreatment at acupoints 'fengchi' and 'waiguan' was used as the test factor.Immunohistochemistry was conducted to determine the protein expression of CB1 receptor and C-fos in trigeminal nucleus caudalis.Relative mRNA expression of the indicated genes in the indicated conditions was quantified using a Step One PlusTM Fast Real-Time PCR system.Results:Effects of electroacupuncture pretreatment on protein and mRNA expression of CB1 in trigeminal nucleus caudalis of migraine rats:CB1 receptor protein and mRNA expression of model group,electroacupuncture pretreatment group and rimonabant antagonist group were significantly upregulated compared with sham operation group(P〈0.05);CB1 receptor protein and mRNA expression of electroacupuncture pretreatment group and rimonabant antagonist group were significantly upregulated compared with model group(P0.05);electroacupuncture pretreatment group had no significant difference in protein and mRNA expression of CB1 receptor compared with rimonabant antagonist group.Effects of electroacupuncture pretreatment on protein and mRNA expression of C-fos in trigeminal nucleus caudalis of migraine rats:C-fos protein and mRNA expression of model group,electroacupuncture pretreatment group and rimonabant antagonist group were significantly upregulated compared with sham operation group(P0.05);C-fos protein and mRNA expression of electroacu
