目的探讨CD133、EGFRvⅢ在人脑胶质瘤组织中的共表达及临床意义。方法选择行手术治疗的脑胶质瘤患者40例,采用免疫组化双重染色法检测其脑胶质瘤组织中的CD133、EGFRvⅢ表达。结果40例患者脑胶质瘤组织中的CD133+细胞、EGFRvⅢ+细胞阳性表达率分别为100%、40%,二者双染阳性表达率为35%。CD133呈棕黄色,位于细胞质和细胞膜上,多数阳性细胞在肿瘤组织中散在分布,部分呈聚集现象;EGFRvⅢ呈深蓝色,主要分布在细胞膜上,细胞质中有少量分布;在肿瘤组织靠近血管区两种阳性细胞存在共聚集现象。结论CD133、EGFRvⅢ在脑胶质瘤组织中的分布具有相关性,在细胞水平上存在共表达,二者可以成为胶质瘤干细胞治疗的分子靶点。
Objective To investigate the co-expression of CD133 and epidermal growth factor receptor variant m ( EGFRvⅢ ) in human glioma tissues and the clinical significance. Methods The expression of CD133 and EGFRv Ⅲ was determined by Double staining of immunohistochemistry in 40 surgical specimens of human glioma tissues. Results The positive rates of CD133 + cells and EGFRvⅢ+ cells in 40 cases were 100% and 40%, respectively. The double staining positive expression rate was 35%. CD133 was claybank in the cytoplasm and cell membrane, the majority of positive cells scattered in the tumor tissues, and some gathered together. EGFRv Ⅲ was dark blue, the mainly distribution was in the membrane, and a few were in the cytoplasm. There was co-aggregation of both positive cells in the tumor tissue near the vascular area. Conclusions There are correlations in the distribution and co-expression at the cellular level between CD133 and EGFRvⅢ in human glioma tissues. Both CD133 and EGFRvⅢ could become the molecule targets for glioma stem cell therapy.
