目的:研究抗癌药物羟基喜树碱(HCPT)新剂型羟基喜树碱脂质体(LHCPT)在Beagle犬体内的毒物代谢动力学,探讨该药物是否在血液中有蓄积作用。方法:采用HPLC-荧光检测法测定全血中LHCPT的浓度。结果:在第1天和连续给药第30天后Cmax和AUC值存在剂量相关性;药物半衰期t1/2不同剂量之间无显著变化(P〉0.05)。AUC、Cmax、t1/2值多次给药与单次给药后比较,同剂量下均未见显著提高(P〉0.05)。结论:LHCPT连续静脉给药后未见血液中有明显蓄积现象。
Objective:To study the toxicokinetics of LHCPT in Beagle dogs by intravenous infusion,and discuss the accumulative effects of LHCPT in blood.Methods:HPLC with fluorescence detection was used to analyze drug concentration in whole blood.Results:Cmax and AUC were increased in a dose-dependent manner both after 1st day and 30th day;and there was no statistical significance in t1/2 of different doses(P 〉 0.05).Compared with single administration with repetition,there were no obvious changes of AUC,Cmax and t1 /2 of the same dose(P 〉 0.05).Conclusion:LHCPT has no significantly accumulative effects after successive intravenous administration.
