目的:探讨高三酰甘油血症(HTG)合并急性心肌梗死(AMI)男性患者载脂蛋白A5(apo A5)基因甲基化对apo A5表达水平的影响。 方法:选取HTG合并AMI男性患者为病例组,选取HTG合并颈动脉粥样硬化(As)男性患者为对照组,每组各18例。用Illumina人类全基因组DNA甲基化450K芯片检测两组患者外周血DNA甲基化变化,用生物信息学方法分析比较两组间apo A5基因的甲基化差异;实时荧光定量聚合酶链反应(RT-PCR)检测apo A5 mRNA表达水平; ELISA法检测血浆apo A5水平。 结果:全基因组甲基化芯片初步筛选结果显示,与对照组相比,病例组apo A5基因表现为高甲基化;RT-PCR和ELISA检测结果显示,病例组apo A5 mRNA及apo A5表达水平低于对照组[(0.037±0.016)vs(1.000±0.093),(156.6±16.8)μg/L vs(197.3±32.3)μg/L,t分别为18.970和2.502, P均〈0.05]。 结论:HTG合并AMI男性患者apo A5的mRNA和apo A5蛋白表达水平均降低与该基因甲基化水平增高有关,apo A5基因甲基化异常可能参与HTG患者的As进展。
Objective:To investigate the effect of the methylation of apolipoprotein A5 ( apo A5 ) gene on the expression level of apo A5 in male patients with hypertriglyceridemia (HTG) and acute myocardial infarction (AMI). Methods:Eighteen male patients with HTG and AMI were included in the case group, while 18 with HTG and carotid atherosclerosis (As) in the control group. The DNA methylation changes in peripheral blood of two groups of patients were detected by the Illumina Human Methylation 450K BeadChip, and the difference of apo A5 methylation between the two groups was compared by the bioinformatic analysis. In addition, the mRNA level of apo A5 and the protein level of plasma apo A5 were determined by real-time fluorescence quantitative PCR (RT-PCR) and ELISA, respectively. Results:The genome wide methylation screening showed that the apo A5 gene was hypermethylated in the case group when compared with the control. The mRNA and protein levels of apo A5 in the case group were significantly lower than that of the control group [(0.037±0.016) vs(1.000±0.093), t=18.970, P〈0.05; (156.6±16.8) μg/L vs (197.3±32.3) μg/L, t=2.502, P〈0.05]. Conclusion:The low mRNA and protein levels of apo A5 in male patients with HTG and AMI may be related to the increased methylation of apo A5 gene, and the abnormal methylation of apo A5 gene may be involved in the progress of atherosclerosis in HTG patients.
