SCCO2体系下,考察了压力,温度,作用时间,含水量,卸压时间对凝胶颗粒完整率和酶的稳定性影响。在不同条件下,影响因素作用的程度不同;快速卸压会引起CO2相交甚至颗粒破裂,卸压越快,海藻酸钙胶体的完整率越低;优化SCCO2体系下酶反应条件为:压力10MPa,温度40℃,相对含水量70%,底物LA与酶质量比为0.2(mg/g),反应时间为1.5h,卸压时间〉2h;进3次循环反应,前2次重复使用具有较好的异构化酶活力。因此,在SCCO2体系下,初步实现了海藻酸钙包埋亚油酸异构酶催化底物LA的转化反应,并可重复使用。
The effects of high - pressure CO2 on the activity of immobilized LA isomerase were studied in every detail. The pressure, temperature, decompression time, content of water and treatment time affected the completeness degree alginate calcium gel and enzymatic activity. The influence degree of parameters varied with experimental conditions. The time of decompression may promoted phase transformation of CO2 and the crack of alginate calcium gel. More rapidly pressure was release lower completeness degree of alginate calcium gel was obtained. The optimized conditions were 10MPa, 40%, treatment time 1.5h, content of water 70%, relatively, and decompression time 〉 2h. The enzymatic isomeric catalysis of reused enzyme was investigated over 3 reused reactions, and only the first two reactions had the better activity of isomeric catalysis. So, the studied results showed that LA isomerase entrapped by embedding medium of calcium alginate gel in the SCCO2 kept efficient and circulatory bioconversion.
