目的发现并评价一种新的高血压小型猪模型,并对其相关靶器官损害情况进行分析。方法监测6、12、18个月龄贵州小型猪（n=5）动脉血压水平,并将其与同月龄家猪（n=5）作对比,测量18月龄时两组随机血糖（GLU）、糖化血红蛋白（HbA1c）、总胆固醇（TC）、甘油三脂（TG）、低密度脂蛋白胆固醇（LDL）、高密度脂蛋白胆固醇（HDL）、肌酐（Scr）、尿素氮水平（BUN）,ELISA法测量其血清肾上腺素（Epinephrine,E）、去甲肾上腺素（Norepinephrine,NE）、多巴胺（Dopamine,DA）及尿香草扁桃酸（Vanilmandelic acid,VMA）水平,放射免疫法测定肾素（Renin）活性、血管紧张素Ⅰ（AngiotensineⅠ）、血管紧张素Ⅱ（AngiotensineⅡ）、醛固酮（Aldosterone）水平,左心室游离壁及肾皮质取材并进行H＆E、MASSON染色。结果与6、12、18同月龄家猪比较,贵州小型猪动脉收缩压及舒张压明显升高（P〈0.01）;18个月龄时贵州小型猪血清TC[（3.11±0.09）mmol/L、TG[（0.62±0.04）mmol/L]、LDL[（1.81±0.05）mmol/L]水平显著高于家猪（均P〈0.05）,而GLU、HbA1c、Scr及BUN水平则无明显差异（均P〉0.05）;贵州小型猪血清E、NE、DA及尿VMA水平分别为[（6968.68±426.10）pg/ml]、[（20.21±0.91）ng/ml]、[（27404.26±4257.96）pg/ml]和[（61.36±2.14）ng/ml]均显著高于家猪（均P〈0.01）,而肾素、血管紧张素Ⅰ、血管紧张素Ⅱ、醛固酮水平则无明显差异（均P〉0.05）;对其靶器官心脏及肾脏HE及Masson染色结果显示,贵州小型猪相对于家猪,左心室游离壁有明显心肌细胞肥大及少量心肌纤维化,心肌细胞横截面面积[（651.05±7.58）μm2]显著高于家猪（P〈0.01）,且肾小球有萎缩及轻微纤维化表现。结论近交系贵州小型猪可成为一种新的动脉高血压动物模型,其交感系统活性活跃,且心脏及肾脏均可形成类似高血压靶器官损害的特征。

Objective To establish and evaluate a novel model of hypertensive mini-pig,and analyze the related target organs damage.Methods The blood pressure（BP）of 6、12、18months Guizhou mini-pigs（n=5）and domestic pigs（n=5）were measured.At the age of 18 months,The plasma glucose,hemoglobin A1c（HbA1c）and lipid profiles including triglycerides,total cholesterol,low-density lipoprotein（LDL）cholesterol and high-density lipoprotein（HDL）cholesterol of both groups were assessed.The plasma concentrations of epinephrine（E）,norepinephrine（NE）,dopamine and urine vanilmandelic acid（VMA）of the two groups were measured using enzyme-linked immunosorbent assay（ELISA）kits.The plasma renin activity and the plasma levels of angiotensin（ANG）Ⅰ,Ⅱ,and aldosterone were assayed by radioimmunoassay.The heart and kidney of two groups were stained with HE,Masson trichrome.ResultsThe BP of 6,12,18 months Guizhou mini-pigs are significantly higher than the same age domestic pigs（P〈0.01）.The serum TC（3.11±0.09）mmol/L、TG（0.62±0.04）mmol/L,LDL（1.81±0.05）mmol/L of the 18 months of Guizhou mini-pigs were higher than that of domestic pig（all P〈0.05）.Moreover,the serum E,NE,DA and urine VMA level of Guizhou mini-pigs were（6968.68±426.10）pg/ml,（20.21±0.91）ng/ml,（27404.26±4257.96）pg/ml and（61.36±2.14）ng/ml,which were significantly higher than domestic pigs（all P〈0.01）.The HE and Masson trichrome stain results show that The mini-pigs demonstrated a significant cardiac hypertrophy and a slight fibrosis and mild renal fibrosis as compared with domestic pigs.Conclusion The Guizhou mini-pig is a novel hypertensive animal model with sympathetic hyperactivity.
