双向电泳技术是当前蛋白质组学研究中的重要技术。设计"双向电泳检测热休克处理对细菌蛋白表达谱的影响"的实验,以PGEX-4T-2大肠杆菌中可溶性总蛋白为研究对象,采用双向电泳技术筛选热休克处理后有差异表达的蛋白。实验获得了稳定性较高、分辨率和重复性较好的电泳图谱,建立了一种适于本科实验教学的双向电泳技术体系。通过该实验,学生可以掌握双向电泳技术的基本原理、实验方法以及蛋白提取的一般思路与方法。
Two-dimensional electrophoresis(2-DE) is an important technology in proteomics research.The experiment of the effect of heat shock treatment on the expression profile of bacterial protein was designed to provide undergraduate an experiment for 2-DE technique.The soluble proteins from Escherichia coli was studied by 2-DE.The higher stability and resolution repeatability 2-DE maps were obtained.Through this experiment,students can master the basic principle,experiment method of 2-DE and the general idea of protein extraction.
