研究了七叶鬼灯檠(Rodgersia aesculifolia)中多酚类化合物最佳提取工艺条件,并采用重氮试剂比色法对总多酚含量进行了测定。在单因素试验的基础上,用正交试验法对总多酚的提取工艺进行优选,选用L9(34)进行正交试验,以总多酚提取率为指标,研究提取溶剂种类、提取溶剂浓度、提取时间、料液比对总多酚提取率的影响。结果表明,最佳提取工艺为:料液比为1∶10的60%甲醇中性溶液,不加超声连续回流提取3次,每次30 min。重氮试剂比色法测得七叶鬼灯檠中总多酚含量为28.32%。提取工艺科学、合理。
The optimum process of extracting total polyphenol from the root of Rodgersia aesculifolia by using univariant method was studied and optimized with orthogonal test, then the content of total polyphenol was monitored by colorimetry of diazotization reagent at 415 nm. Experiments were conducted by using L9 (34) orthogonal design with extractive rate of total polyphenol as the index, the influence of extracting solvent, the concentration of extracting solvent, extracting time and the ratio of solid to liquid were determined. The optimum extracting condition of total polyphenol was that the material was refluxed in 10 times (m/v) 60% of neutral methanol solution (v/v) for 30min with 3 times without ultrasonic both. The content of total polyphenol from R. aesculifolia was 28.32% The results indicated that the optimum extraction conditions of total polyphenol was rational.
