目的探讨突触后致密体蛋白95(postsynaptic density protein 95,PSD-95)在大鼠前额叶皮质发育中的表达变化。方法采用real-time PCR和Western blot分别测定出生后第1,7,14,21,30,50天的Wistar大鼠前额叶皮质PSD-95的mRNA及蛋白水平的表达。结果大鼠在出生后不同时间,其前额叶皮质PSD-95 mRNA及其蛋白表达量有一定的特点。在出生后第1-7天,PSD-95 mRNA及其蛋白表达水平均较弱;出生后第14-30天,PSD-95 mRNA及其蛋白表达水平明显增强(P〈0.01);出生后第50天继续表达在较高水平(P〈0.05),但较出生后第14-30天已有所减弱。结论 PSD-95在前额叶皮质中的表达变化具有时间特异性,此模式可能与突触的发育及成熟变化密切相关。
Objective To investigate the changes of expression of postsynaptic density protein-95 ( PSD-95 ) in the prefrontal cortex of rat at postnatal stage. Methods Real-time PCR and Western blot were used to detect the expression of PSD-95 mRNA and protein respectively at postnatal day 1, 7, 14, 21, 30 and 50. Results The expression of PSD-95 mRNA and protein in prefrontal cortex showed some characteristics during the postnatal development. At postnatal day 1 -7 ,the expression levels of PSD-95 mRNA and protein were weak. At day 14 -30 ,the expression levels of PSD-95 mRNA and protein were significantly increased compared with that at postnatal day 1 (P 〈 0.01 ). At postnatal day 50, the expression levels of PSD-95 mRNA and protein were expressed in a high level( P 〈 0.05 ), but they were weaker than those at postnatal day 14 - 30. Conclusion The expression of PSD-95 shows a special temporal distribution pattern, and PSD-95 may participate in the development and maturation of synapse.
