分别构建RNAi(RNA interference,RNAi)载体干扰家蚕细胞BmN中Argonaute1(Ago1)和Argonaute2(Ago2)基因的表达。首先利用PFBDM和pBac[A3-EGFPafm]载体构建Ago1和Ago2基因的RNA干涉载体,通过转染家蚕细胞发现,Ago1和Ago2基因表达明显下降。并观察细胞形态发现,转染RNAi载体96h后,家蚕BmN细胞逐渐失去正常的梭形状态,体积变大,形态变圆。该研究为进一步研究家蚕中small RNA的产生及作用机制提供了较好的基础。
To silence the gene expression of Argonautel (Agol) and Argonaute2 (Ago2) in silk- worm Bran cells, the RNA interference (RNAi) vectors were constructed and transfected into BmN cells. The results showed that the expressions of Ago1 and Ago2 were decreased and the cells turned rounder and bigger at 96 h post-transfection.
