淫羊藿苷通过降低MMP-9抗自发性高血压大鼠心室重构

目的 探讨高血压对大鼠有机阴离子转运多肽(organic anion transporting polypeptides,OATPs/Oatps)及多药耐药相关蛋白(multidrug resistance-associated protein,Mrps)mRNA水平的影响。方法 14周龄的雄性自发性高血压大鼠(Spontaneous Hypertension Rat,SHR)为SHR组(SHR,n=10),同源正常的京都大鼠(Wistar-Kyoto rat,WKY)为WKY组(WKY,n=10)。常规饲养12周。每两周记录各组大鼠体重变化、Kent Scientific CODA系统检测大鼠血压变化。Real time RT-PCR检测大鼠肝脏组织中的主要摄取性转运体Oatp1a1、Oatp1a4、Oatp1a5、Oatp1b2及主要外排型转运体Mrp1、Mrp2、Mrp3、Mrp4 mRNA的水平。结果 与WKY组相比,SHR组大鼠血压明显上升(P〈0.05),SHR组大鼠肝脏转运体Oatp1a1、Oatp1a4、Oatp1a5、Oatp1b2、Mrp2及Mrp4 mRNA的表达量显著增高(P〈0.01),而Mrp3 mRNA的表达量显著降低(P〈0.01),Mrp1 mRNA的表达量无明显变化(P〉0.05)。4种OATP中,Oatp1b2 mRNA在WKY(32.77%of f'-actin)及SHR(79.59%of f'-actin)中表达均最高,Oatpla5 mRNA在WKY(0.0033%of f'-actin)及SHR(0.009 2%of f'-actin)中表达均最低。4种Mrp中Mrp2 mRNA在WKY(22.46%of f'-actin)及SHR(61.04%of f'-actin)中表达均最高。结论 高血压对大鼠肝脏主要摄取性转运体Oatp1a1、Oatp1a4、Oatp1a5、Oatp1b2及主要外排型转运体Mrp2、Mrp3、Mrp4 mRNA的表达有影响,而对Mrp1 mRNA的表达无影响。

英文摘要:

Objective To investigate the effects of spontaneous hypertension on the mRNA expression of organic anion transporting polypeptides (Oatps) and muhidrug resistance - associated proteins (Mrps) of hepatic tissue in spontaneously hypertensive rats (SHR). Methods Fourteen weeks old male SHR were the SHR group ( SHR, n = 10). The same aged male homologous Kyoto rats (WKY) were the WKY group (WKY,n : 10). Rats were routine fed for 12 weeks. The mRNA expression of the major uptake transporter Oatplal, Oatpla4, Oatpla5 and Oatplb2 as well as the main efflux transporters Mrpl, Mrp2, Mrp3 and Mrp4 were detected by Real time RT- PCR. The weight change of rats in each group were recorded every two weeks. Blood pressure change was detec- ted in rats with Scientific CODA Kent system. Results Compared with the WKY group, the blood pressure in SHR group was significantly increased ( P 〈 0.05 ). The mRNA expression of Oatpl al, Oatpl a4, Oatpl aS, Oatpl b2, Mrp2 and Mrp4 were up- regulated (P 〈0.01 ) and the mRNA expression of Mrp3 was down- regulated (P 〈 0.01 ) in SHR group. However,the mRNA expression of Mrpl did not ehanged significantly (P 〉0.05). Among four Oatps,the mRNA expression of Oatplb2 was the highest (32.77% or 79.59% of 13 - actin) in WKY or SHR groups,and Oatpla5 was the lowest (0. 0033% or 0. 0092% of 13 -actin) in WKY or SHR groups. Among four Mrps,the mRNA expression of Mrp2 was the highest (22.46% or 61.04% of 13 -aetin) in WKY or SHR groups. Conclusion The mRNA expression of Oatpl al, Oatpl a4, Oatpl a5, Oatpl b2, Mrp2, Mrp3 and Mrp4 in the main liver transporters of rats were significantly affected by hypertension. However, hypertension has no significant effect on the mRNA expression of Mrpl.
