目的:研究珠子参总皂苷对对四氯化碳所致大鼠慢性肝损伤的保护作用,并探讨其作用机制。方法:采用40%的CCl4橄榄油剂灌胃制备慢性肝损伤大鼠模型。分别采用不同浓度的珠子参总皂苷进行干预,9周后检测大鼠肝、脾脏器指数,血清ALT、AST浓度和TP、ALP、ALB含量,肝组织中MDA、SOD、T-AOC、GSH-Px、Hyp的含量。结果:珠子参总皂苷(60、120、240 mg/kg)灌胃给药能显著降低CCl4致慢性肝损伤大鼠肝脏、脾脏指数;降低血清中异常升高的ALT、AST;提高血清中降低的TP含量,降低血清中升高的ALP含量,降低肝组织中MDA水平,同时升高其低下的T-AOC、GSH-Px水平;降低Hyp含量。结论:珠子参总皂苷对大鼠实验性肝损伤具有一定程度的保护作用,其机制可能与抑制脂质过氧化反应、减少自由基生成、提高组织抗氧化能力、促进蛋白质合成有关。
Objective: To study the protective effect of total saponins of Panacis majoris rhizoma against CCl4 induced chronic hepatic injury and investigate its mechanism. Methods: The hepatic fibrosis rat model was developed by gavage of 40% CCl4 and rats were treated with total saponins of Panacis majoris rhizoma separately at 240 mg / kg,120 mg / kg and 60 mg / kg. Liver index,pleen index,the serums levels of ALT and AST,alkaline phosphatase( ALP),total protein( TP) and albumin( ALB) levels were measured. Results: Total saponins of Panacis majoris rhizoma( 60,120,240 mg / kg) could significantly reduce the CCl4 to chronic liver injury rat liver spleen index,reduce the abnormally high serum level of ALT and AST; increase serum level of TP,and reduce the level of ALP and malondialdehyde. The degeneration level of T-AOC,GSH-Px were markedly increased in the model group. Conclusion: Total saponins of Panacis majoris rhizoma can protect experimental hepatic injury and prevent the progression of hepatic fibrosis in rats. The mechanism maybe related to the inhibition of lipid peroxidation,reducing the creation of free radicals,improvement antioxidant capacity and romoting the synthesis of proteins.
