RT-PCR、Western印迹、免疫荧光3种方法检测大鼠脂肪中富含半胱氨酸的酸性分泌蛋白(SPARC)mRNA和蛋白的表达。胰岛素抵抗大鼠脂肪中SPARC mRNA和蛋白的表达高于普通大鼠(均P〈0.01),SPARC在胰岛素抵抗发展中可能起到负反馈调节的作用。
[ Summary ] RT-PCR, Western blot, and immunofluorescence were used to examine secreted protein acidic and rich in cysteine ( SPARC ) mRNA and protein expression in adipose tissue of rats. The results showed that the expression levels of SPARC mRNA and protein in adipose tissue of insulin resistant rats were higher than those of the normal ones( all P〈0.01 ). SPARC may play a role in the negative feedback regulation duning the development of insulin resistance.
