目的观察姜黄素对人骨肉瘤细胞系Saos-2增殖和凋亡的影响及其对Fas蛋白表达的调控。方法体外培养人骨肉瘤细胞系Saos-2,MTT法检测不同浓度姜黄素对骨肉瘤细胞系Saos-2的生长抑制作用,Annexin V/PI染色,流式细胞术检测细胞凋亡,Hoechst 33258染色,显微镜下观察细胞凋亡的形态学变化,Western blot法检测Saos-2细胞Fas蛋白表达情况。结果 MTT检测显示姜黄素可抑制人骨肉瘤Saos-2细胞增殖,并具有时间和剂量依赖性,在姜黄素浓度为20μmol·L-1时,对细胞生长的抑制作用明显增强;流式细胞检测显示姜黄素可明显诱导骨肉瘤细胞凋亡;Hoechst33258染色可见凋亡小体;Western blot分析显示经姜黄素作用后的骨肉瘤细胞系Saos-2表达Fas较用药前明显增强。结论姜黄素对骨肉瘤细胞系Saos-2的增殖有抑制作用,并可诱导其发生凋亡,其可能通过上调Fas的表达,启动细胞凋亡程序,发挥其抗肿瘤作用。
Aim To investigate the effect of curcuma on the growth inhibition,apoptosis and the expression of Fas protein in human osteosarcoma cell line Saos-2.Methods Osteosarcoma cell line Saos-2 was cultured in vitro.Cell proliferation was detected by Methyl thiazolyl tetrazolium(MTT).Stained by Annexin V and PI the cells were observed by flow cytometry(FCM) for apoptosis.Hoechst 33258 was used to detect the morphological change of apoptotic cells.The protein expression of Fas in Saos-2 was observed by westernblot.Results The proliferation of human osteosarcoma cell line Saos-2 could be inhibited and apoptosis could be induced by curcuma.The inhibition rate and apoptosis rate were different with the different concentrations.The inhibition rate and apoptosis rate of Saos-2 cells rose significantly when the concentration of curcuma was over 20 μmol·L-1.Apoptotic body could be observed by Hoechst 33258 dying.The Fas protein expression of human osteosarcoma cell line Saos-2 increased obviously by using curcuma.Conclusions Curcuma could inhibit the proliferation and induce apoptosis of human osteosarcoma cell line Saos-2.The therapeutic efficacy might be induced by the apoptosis procedure initiated through up-regulating the expression of Fas.
