为了研究敌敌畏(dichlorvos,DDVP)对人肾细胞的毒性效应。应用MTT比色法检测敌敌畏对人肾细胞系293T细胞增殖的影响,流式细胞仪检测293T细胞的凋亡情况,荧光二氯荧光素二乙酸(DCF-DA)测定细胞内的活性氧(reactive oxidative species,ROS)水平。结果表明:用浓度为0.5mmol/L、1mmol/L、5mmol/L、10mmol/L、15mmol/L和20mmol/L的敌敌畏分别处理293T细胞1h后发现敌敌畏对293T的生长抑制具有剂量效应,计算得到1h的IC50值为10.406mmol/L;不同浓度敌敌畏(0.5mmol/L,5mmol/L,10mmol/L)处理1h293T细胞发现细胞产生明显的凋亡现象,随着处理浓度的增加,细胞的凋亡率升高,检测细胞内活性氧水平发现细胞内活性氧水平也随之升高。结果提示敌敌畏能够通过凋亡途径抑制293T细胞的增殖,其作用机制可能是通过增加293T细胞内活性氧水平,促进细胞凋亡,进而抑制细胞增殖。
To explore the toxicological effect of dichlorvos (DDVP) on kidney cell, the proliferation of kidney 293T cells were measured by MTT method, apoptosis of cells was detected by flow cytometry, and the level of reactive oxidative species (ROS) in cells was determined by DCF-DA. The results showed that the cell growth of 293T that treated with 0.5 mmol/L, 1 mmol/L, 5 mmol/L, 10 mmol/L, 15 mmol/L, 20 mmol/L DDVP for 1 h was inhibited, and the ICs0 of 1 h was 10.406 mmol/L. Different dose DDVP (0.5 mmol/L, 5 mmol/L, 10 mmol/L) could induce obvious apoptosis phenomenon on 293T after 1 h, and the apoptosis rate and the level of ROS in 293T went up with the increase dose of DDVP. These results indicated that DDVP could effectively inhibit the cell growth via apoptosis pathway in 293T cells, whose mechanism might be realized by increasing the level of ROS in 293T cells, promoting cell apoptosis, thereby inhibiting cell proliferation.
