目的研究5-羟色胺转运体(5-HTT)基因LPR和VNTR多态性与重性抑郁障碍(MDD)中睡眠障碍的关系。方法选取441例中国汉族MDD患者作为研究对象,运用聚合酶链反应技术(PCR)检测5-HTrLHR和5-HTTVNTR基因多态性。根据汉密顿抑郁量表(HAMD)评定患者的睡眠障碍,采用UNPHASED软件进行数量性状分析。结果①5-HTYLPR多态性的等位基因与MDD患者HAMD的睡眠障碍因子相关,即不同等位基因组的入睡困难、睡眠不深、早醒、睡眠因子评分差异均有统计学意义(X^2为20.51、21.50、16.85、30.13,P均小于0.05),携带S等位基因的MDD患者较易伴发睡眠障碍。5-HTFLPR多态性的基因型与MDD患者入睡困难、睡眠不深、早醒及睡眠因子评分相关(X^2为19.48、20.46、19.98、29.38,P均小于0.05),S/S基因型的MDD患者更易伴发睡眠障碍。②5.HTTVNTR多态性等位基因、基因型与MDD睡眠障碍因子的数量性状分析结果均为阴性(P〉0.05)。③两个多态性的不同单倍型(Stin2.12-L、Stin2.12-S、Stin2.10-L、Stin2.10-S)问的上述睡眠障碍评分差异均有统计学意义(X^2为21.32、34.82、18.68、34.55,P均小于0.05)。结论在中国汉族人群中,5-HTTLPR多态性可能与抑郁症睡眠障碍有关。
Objective To study the relationship between 5-HTT gene polymorphisms (5-HTTLPR and 5-HTTVNTR) and items scores of sleep disorder on MDD. Methods We selected 441 Chinese Han patients with major depressive disorder as subjects. The genotypes of two polymorphisms (5-HTTLPR and 5-HTTVNTR) in 5-HTT gene were determined by using polymerase chain reaction. The depression states were assessed with Hamilton Depression Rating Scale (HAMD). Sleep disorder as the quantitative traits was analyzed by the QTPHASE modules of UNPHASED software. Results The quantitative trait test revealed that the allele and the genotype of 5-HTI'LPR, but not 5-HTFVNTR polymorphism had strong genetic relationship with factor and items scores of sleep disorder on MDD ( X^2 = 20. 51,21.50,16. 85,30. 13,19.48, 20. 46,19.98,29. 38, P 〈 0.05 ). There was a strong relationship between the haplotype of 5-HTr gene and factor and items scores of sleep disorder on MDD (X^2 =21.32,34. 82,18. 68,34. 55 ,P 〈0. 05). Conclusions 5-HTTLPR is likely to associate with the factor and items scores of sleep disorder on MDD in Chinese Han Population.
