为探讨人脐静脉内皮细胞(HUVEC)中小凹蛋白-1与细胞外钙受体表达及定位,为进一步功能研究提供理论依据,采用Western—blot和Rt—PCR及免疫荧光技术检测HUVEC中小凹蛋白-1与细胞外钙受体mRNA和蛋白表达及定位。结果显示:(1)形态学观察和免疫细胞化学法测Ⅷ因子抗原,95%以上的细胞呈阳性着色,证实细胞为HUVEC。(2)RT-PCR检测到HUVEC中459bp和260bpmRNA表达,Western—blot测到HUVEC中22KD和150—160KD蛋白表达。(3)免疫荧光检测到小凹蛋白-1与细胞外钙受体定位于人脐静脉内皮细胞膜。由此可知,人的HUVEC中有小凹蛋白-1与细胞外钙受体表达,两者是否共定位及其功能有待进一步研究。
To investigate the expression of caveolin-1 and extracellular calcium receptor (CAR) and localization in human umbilical vein endothelial cells(HUVECs),which provide a theoretical basis for further colocalization and functional studies. Methods:The expressions of caveolin-1 and CaR mRNA and protein in HUVECs were detected by Western-blot and Rt-PCR, and their localizations were investigated by immunofluorescence. Results: (1) We identified HUVECs in which over 95% of cells were positive staining by morphology and immunocytochemistry measuring Ⅷ factor antigen. (2) The expression of 459bp, 260bpmRNA and 22KD, 150-160KD protein were detected in HUVEC by Western-blot detected and RT- PCR,respectively. (3)Caveolin -1 and CaR were localization in HUVEC membrane by immunofluorescence. Conclusions: Our observations demonstrated that there were caveolin -1 and CaR in HUVECs,and they were localization in the cell membrane. Whether they were co-localization and the relation between the expression and its function should be further studied.
