目的:观察苯妥英钠和盐酸米诺环素复合膜对实验性大鼠牙周骨开窗缺损再生过程中骨形成情况及BMP-2表达水平的影响。方法:制备含有苯妥英钠和盐酸米诺环素的胶原膜和不含药物的空白膜,分别置于30只Wistar大鼠磨牙区牙周骨开窗缺损内,每组15只。分别于术后10d(n=7)和21d(n=8)处死大鼠,取术区组织。HE染色镜下观察缺损区内新生骨样组织形成情况;用免疫组化法检测术后10d和21d缺损区内BMP-2的表达情况,比较各组BMP-2平均积分光密度值。结果:术后10d时,实验组缺损区内被大量成纤维细胞、炎症细胞等占据,多数细胞BMP-2表达呈强阳性,对照组缺损区内细胞量较少,BMP-2表达较弱,二者有显著的统计学差异;术后21d,实验组内见大量片状成骨,BMP-2表达强度较10d时减弱,对照组缺损内有少量散在成骨,BMP-2表达较10d时增强,但二者BMP-2表达不存在显著性差异。结论:苯妥英钠和盐酸米诺环素复合膜可能增强大鼠牙周骨组织再生过程中局部缺损区内BMP-2的表达。
AIM: To investigate the effect of sodium phenytoin (PHT) and minocycline hydrochloride (MC) compound on the healing of the defect and the expression of bone morphogenetic protein - 2 ( BMP - 2 ) during the periodontal bone regeneration. METHODS: A compound of PHT and MC with collagen sponge was prepared using routine method. Following surgery to create a bony window on the buccal aspect of the mandibular molar root, 30 male Wistar rats were divided into two groups randomly, treated with test sponge (n = 15 ) and control sponge (n = 15 ) respectively. In each group, animals were sacrificed after 10( n = 7 ) or 21 ( n = 8 ) days and then the mandibles were processed for histological examination and immunohistochemical staining. The optical density (OD) of BMP -2 was analysed and compared between groups. RESUILTS: At day 10, the defect area in the test group was filled with fibroblast cells and inflammatory cells while relatively less cells were seen in the control group. BMP-2 expression was seen in all these cells. The OD of BMP -2 in the test group was statistically greater than that in the control group. At day 21, large area of newly formed lamellar bone was seen in the test group while less bone in the control one. No statistical difference was found in BMP - 2 expression between these two groups. CONCLUSION:The compound of PHT and MIN is able to enhance the expression of BMP -2 during the regeneration of periodontal alveolar bone in rat mandibles.
