目的探讨肼苯哒嗪(HYD)联合硝酸异山梨酯(ISDN)对心力衰竭大鼠内皮功能及氧化应激水平的影响。方法利用阿霉素诱导大鼠心力衰竭模型,成模后分为安慰剂对照组(给予生理盐水)、培哚普利(2 mg.kg-1.d-1)+比索洛尔(5 mg.kg-1.d-1)组及培哚普利(2 mg.kg-1.d-1)+比索洛尔(5 mg.kg-1.d-1)+HYD(7.2 mg.kg-1.d-1)+ISDN(6 mg.kg-1.d-1)组。药物干预6周后,取心肌组织测定心肌组织内皮型一氧化氮合酶(eNOS)和内皮素1(ET-1)的表达,同时测定丙二醛(MDA)和超氧化物歧化酶(SOD)水平。结果 HYD+ISDN并不能增加心肌组织eNOS含量,对心肌组织内eNOS及ET-1的表达均无显著影响;HYD+ISDN可显著降低心肌组织MDA含量,同时使心肌组织SOD活性增高。结论 HYD+ISDN可改善心力衰竭大鼠体内氧化应激状态。
Objective To investigate effect on endothelial function and oxidative stress in heart failure rats with hydralazine(HYD) combined isosorbide dinitrate(ISDN).Methods We created a heart failure rat model with adriamycin,then divided the heart failure rats into three groups.A group were treated with saline.A group were treated with perindopri and bisoprolol.One group were treated with perindopril + bisoprolol + HYD + ISDN.The levels of MDA,SOD,eNOS,and ET-1 expression from the heart failure rats′ myocardial tissue were determinated after 6 weeks.Results HYD + ISDN didn′t increase myocardial eNOS and had not significant effect on the expression of eNOS and ET-1.HYD + ISDN could reduce myocardial MDA and increase myocardial SOD significantly.Conclusion HYD + ISDN can alleviate the oxidative stress in heart failure rats.
