目的:观察四逆散和六味地黄丸含药血清对体外培养海马神经干细胞(NSCs)的经典Wnt信号通路及非经典Wnt信号通路的影响,从NSCs的增殖与分化机制探讨肝肾"藏泄互用"关系的内涵。方法:分离新生24h内SD大鼠海马NSCs进行体外传代培养,采用巢蛋白免疫荧光染色鉴定NSCs。将第3代NSCs分别采用10%的正常血清、六味地黄丸低、高剂量(2.08、2.66g/mL)含药血清、四逆散低、高剂量(1.33、2.66g/mL)含药血清培养7d。观察各组细胞的增殖能力,检测Wnt1、β-catenin、Wnt5a、JNK蛋白的相对表达量。结果:①六味地黄丸高、低剂量含药血清组的细胞增殖能力最强,与其他各组比较均有统计学意义(P〈0.05,P〈0.01)。②Wnt1、β-catenin表达量在六味地黄丸高、低剂量及四逆散高、低剂量含药血清组均明显高于正常血清对照组(P〈0.01),其中Wnt1的表达量在六味地黄丸高、低剂量含药血清组的表达量最高。Wnt5a、JNK的表达量在六味地黄丸高、低剂量含药血清组均低于其他各组(P〈0.01),其中Wnt5a在四逆散高、低剂量含药血清组表达均高于其他各组(P〈0.01);JNK在四逆散低剂量含药血清组表达量最高,与其他各组比较均有统计学意义(P〈0.01)。结论:在NSCs层面上,肾主藏精主要表现为促进NSCs的增殖和分化;肝主疏泄主要表现为促进NSCs的迁移与分化;而肝肾"藏泄互用"的关系实质内涵可体现为对NSCs的增殖、迁移及分化的协调平衡调控作用。
Objective: To observe the effects of the serum containing Sini Powder and Liuwei Rehumannia Pill on the classic Wnt signal pathway and the non-classic Wnt signal pathway of the hippocampal neural stem cells (NSCs) in vitro. The connotation of interdependence between storing and discharging of liver and kidney was interpreted with the proliferation and the differentiation of the hippocampal NSCs. Methods: The NSCs isolated from the hippocampus of the neonatal SD rats within 24h of birth were cultured in vitro and identified by the nestin immunohistochemical fluorescence staining For the NSCs of 3rd generation, it was cultured respectively with 10% normal serum, the low- and high-doses of serum containing Sini Powder the low- and high-dose of serum containing Liuwei Rehumannia Pill. After 7 days, the proliferative ability of the NSCs was observed in each group and the relative expression quantities of Wntl, β-catenin, Wnt5a and JNK were detected. Results: ①The proliferative capacities of the high- and the low-dose of Liuwei Rehumannia Pill were strongest. There was the statistical significance between the former two groups and the other groups (P〈0.05, P〈0.01). ②The expression quantity of Wntl and β-catenin in the groups of the high and the low-dose of the serum containing Sini Powder and Liuwei Rehumannia Pill was higher obviously than that in the normal serum group (P〈0.01). Among the groups, the expression quantity of Wntl in the groups of the high- and the low- dose of the serum containing Liuwei Rehumannia Pill was highest. The expression quantity of Wnt5a and JNK in the groups of the high- and the low-dose of the serum containing Liuwei Rehumannia Pill was lower than that in the other serum groups (P〈0.01).
