骨是动态的活性组织,通过由骨吸收和骨形成所组成的骨重建来维持自身的平衡及结构的完整性。核因子kB受体活化因子--kB受体活化因子配体--骨保护蛋白通路是骨吸收和骨形成发生耦联的关键通路,其对于维持骨重建的平衡、成骨细胞的分化以及调控破骨细胞生成等过程起着重要的作用。此外,此通路的深入研究对于骨质疏松症、心血管疾病的临床治疗具有重要的指导意义。本文就RANK-RANKL-OPG信号通路在骨发生和吸收中的分子作用机制作一综述。
Bone is a dynamic organization which sustains its balance and structural integrity through bone remodeling that composed of bone resorption and bone formation.Receptor activator of nuclear factor kB(RANK)-receptor activator of nuclear factor kB ligand(RANKL)-osteoprotegerin(OPG)has major effects on bone remodeling,especially in the osteoblast lineage to regulate the differentiation and activity of osteoclasts.In addition,the further research of this pathway will have important guiding significance for the clinical treatment of osteoporosis and cardiovascular disease.This article aims to explain the RANK-RANKL-OPG signaling pathway's molecular mechanism in bone.
