目的探讨周期性压力对大鼠软骨细胞细胞外信号调节激酶(ERK)1和ERK2的影响。方法将软骨细胞置于压力为150kPa,频率为0.1Hz的周期性应力系统中。分为四组:A组不加压作为对照;B、C和D组分别加压0.5、1和2h。用细胞免疫荧光法观察磷酸化ERK1和ERK2(pERK1和pERK2)在软骨细胞内的定位表达,同时用Westernblot法检测ERK1和ERK2在软骨细胞内的磷酸化。结果加压组的软骨细胞内pERK1和pERK2的荧光强度较A组明显增强;C组的荧光强度最大。pERK1和pERK2在B、C、D组内的表达较A组增高(P〈0.01)。结论适当的压力刺激可导致软骨细胞内pERK1和pERK2表达增高,ERK1和ERK2可能在软骨细胞将压力信号转导为生物化学信号过程中起重要作用。
Objective To investigate the effect of cyclic pressure on extracellular signal-regulated kinases 1 and 2(ERK1,ERK2) in rat chondrocytes.Methods The rat chondrocytes were divided into four groups.Groups of B,C and D were mechanically stimulated with 150 kPa,0.1 Hz system for 0.5 h,1.0 h and 2.0 h,respectively.Group A was not treated with cyclic pressure as the control.The phosphorylation of ERK1 and ERK2(pERK1 and pERK2) was detected by immunofluorescence and Western blot.Results The immunofluorescence intensity of pERK1 and pERK2 increased after loading in groups of B,C and D,of which the increase of group C was the greatest.The expressions of pERK1 and pERK2 following cyclic pressure are higher in groups of B,C and D than those in group A(P〈0.01).Conclusion ERK1 and ERK2 are activated following cyclic pressure in rat chondrocytes,in which ERK1 and ERK2 may play an important role in machanical signal transduction of chondrocytes.
