目的 研究氟化物对成纤维细胞Ⅰ型胶原表达和成骨结节形成的影响。方法 将成纤维细胞分为对照组和6个加氟组,氟质量浓度分别为0.0001、0.0010、0.1000、1.0000、10.0000、20.0000mg/L。应用RT—PCR法和ELISA法检测各组成纤维细胞Ⅰ型胶原tuRNA和蛋白的表达。同时应用茜素红染色法观察矿化诱导液对成纤维细胞成骨结节形成的作用。结果 染氟2h的0.0001、0.0010、0.1000mg/L组,染氟4h的0.1000mg/L组.染氟24h的1.0000、10.0000、20.0000mg/L组及染氟72h的10.0000、20.0000mg/L组成纤维细胞Ⅰ型胶原蛋白水平明显升高:染氟48h成纤维细胞Ⅰ型胶原tuRNA的表达增强趋势明显。染氟10d,受矿化诱导液诱导的成纤维细胞出现集中现象。并形成较多数量、大小不等的成骨结节,有明显的钙盐沉积。结论 氟化物可明显促进成纤维细胞成骨表型表达和成骨活动增强。
Objective To study the effects of fluoride(F^-) on the expression of collagen type Ⅰ of fibroblasts (Col Ⅰ) and the formation of mineralized nodule in fibroblast. Methods Fibroblasts were divided into control group and six fluoride groups(0.0001, 0.0010, 0.1000, 1.0000, 10.0000, 20.0000 mg/L F^-). The levels of Col Ⅰ mRNA and protein at certain time were measured by means of RT-PCR and ELISA method, the formation of mineralized nodule induced by ascorbic acid and 13-glycerlphosphate was observed. Results The content of Col Ⅰ protein increased obviously in when exposed to 0.0001, 0.0010 and 0.1000 mg/L F^- at 2 h, to 0.1000 mg/L F^- at 4 h, to 1.0000, 10.0000 and 20.0000 mg/L F^- at 24 h and 10.0000 and to 20.0000 mg/L F^- at 72 h. The increasing tendency of Col Ⅰ mRNA was found after exposing to fluoride for 48 h. The congregation of fibroblasts and the sporadic formation of mineralized nodules were observed in the groups treated with fluoride for 10 days. Conclusions Fluoride plays an important role in enhancing the expression of osteoblastic phenotype in fibroblast and increasing the osteogenesis ability of fibroblast.
