以壳低聚糖与氯乙酰氯反应制备出中间产物2-氯乙酰壳低聚糖,中间产物再与吡唑反应得到2-吡唑酰壳低聚糖.通过FT-IR、1H-NMR、热重分析、溶解度实验对产物进行了结构和性能表征,并测定了产物对褐腐病菌和灰霉病菌的抑菌活性.结果表明:目标产物结构正确,热稳定性略低于壳低聚糖;在水中的溶解度为3 mg·mL-1;抑菌研究表明,产物的抑菌活性随浓度的增大而增大,对褐腐病菌菌丝生长抑制的IC50值为0.33 mg·mL-1,对灰霉病菌菌丝生长抑制的IC50为0.45 mg·mL-1,抑菌活性优于壳低聚糖.
2-chloracetyl oligo-chitosan was obtained by the reaction of oligo-chitosan and chloroacetyl chloride, and then 2-pyrazole acylation oligo-chitosan was synthesized by the reaction of 2-chloracetyl oligo-chitosan and pyrazole. The performance and structure of the product were characterized by FT-IR, 1H-NMR, thermogravimetry (TG), solubility and antibacterial activity analysis. The results showed that the derivative is the target product with its thermal stability inferior to oligo-chitosan. The solubility of derivate is 3 mg-mL-1 in water. The results indicate that the antimicrobial activitiy of the oligo-chitosan derivative increases with the increase of its concentration. It was also found that the antimicrobial activities of the derivative are stronger than those of oligo-chitosan, and the 50% inhibition concentrations of 2-pyrazole acylation oligo-chitosan for the growth of Moniliniafructicola and Botrytis cinerea are 0.33 and 0.45 mg.mL-1, respectively.
