目的探讨四生汤对单次60Co辐射小鼠保护作用及其作用机制。方法建立60Co辐射损伤小鼠模型。实验设对照组,60Co模型组,小、中、大剂量四生汤组。60Co照射后不同时间采血进行血常规、血清总抗氧化能力(T-AOC)、超氧化物歧化酶(SOD)及丙二醛(MDA)检测;2周后处死小鼠计算脾脏系数、胸腺系数,检测骨髓有核细胞数。结果在6Gy60Co辐射下,与模型组相比,四生汤能以剂量依赖方式升高小鼠脾、胸腺系数(P〈0.01);高剂量的四生汤能显著升高外周血白细胞、红细胞、血红蛋白和血小板含量及骨髓有核细胞数(P〈0.01);并能剂量依赖性地提高T-AOC和SOD活性(P〈0.01)。结论四生汤能减轻辐射对外周血毒性,并能够抑制受辐射小鼠的氧化损伤。
Objective To observe the protective effect and action mechanism of Sisheng Decoction on 60Co radiation-induced injury of mice.Methods Kun-Ming mice were divided into control,model,and low,medium,high Sisheng Decoction dosage groups after 60Co irradiation.Spleen and thymus indexes and all kinds of blood cells were counted.Oxidative stress was estimated using SOD,MDA and TAOC in mice following 60Co irradiation.Micro-nucleated cells of bone marrow were calculated.Results Sisheng Decoction increased the spleen and thymus indexes and serum SOD and TAOC activities,while decreased the MDA production in mice 60Co irradiation compared to model group in a dose-dependent manner,showing that Sisheng Decoction can increase the number of whole blood cells and micro-nucleated bone marrow cells in mice after 60Co irradiation.Conclusion Sisheng Decoction can protect bone marrow hematopoietic function of mice,reduce toxicity of 60Co irradiation to peripheral blood,and inhibit 60Co irradiation-induced oxidative damage in mice.
