目的:一通过检测线粒体膜电位的变化,探讨巨细胞病毒对线粒体膜电位的影响以及药物对线粒体膜电位的保护作用。方法:制备细胞飞片,用共聚焦显微镜检测正常对照组、病毒组、金叶败毒组和更昔洛韦组的细胞线粒体膜电位。结果:正常对照组各时相线粒体膜电位无明显变化。病毒组12h线粒体膜电位增高,48h线粒体膜电位开始降低,72h膜电位明显降低,部分细胞脱落。金叶败毒组和更昔洛韦组与正常细胞组相比差异无显著性,与病毒对照组比较24h、48h和72h3个时相差异具有显著性。结论:豚鼠巨细胞病毒感染可以导致线粒体膜电位降低,诱导细胞凋亡;金叶败毒和更昔洛韦对豚鼠巨细胞病毒所诱导的线粒体膜电位损伤有保护作用。
Objective: To investigate the change of mitochondrion membrane potential, to study the effect of cytomegalvirus (CMV) and the protection of medicines, Methods: The cells on coverslip were cultured toobserve the mitochondrion -membrane potential of control group, CMV group, JYBD group and ganciclovir group by laser scanning apoptosis microscopy. Results: The mitochondrion mem- brane potential of normal controlled group was obviously different. The mitochondrion membrane potential of CMV group was improved at 12 h; it was degraded at 48 h; obviously decreased at 72 h. In JYBD and GCV groups, compare with the control group the mitochondfion membrane potential was't significant, but compare with the CMV group, mitochondrion membrane potentialwas significant at 24, 48 and 72 h. Conclusion: The infection of CMV induced the mitochondfion membrane potential has no significant decrease and cell apoptosis. JYBD and GCV can protect the cell by inhibiting the mitochondrlon membrane potential damage from CMV infection.
