目的:建立混合血清孵育的肺腺癌组织双向蛋白印迹图谱。方法:收集2例肺腺癌患者的手术标本,提取癌组织可溶性总蛋白并双向凝胶电泳分离,将蛋白转移到硝酸纤维素膜上,然后分别与4例肺腺癌患者的混合血清和4例良性肺部疾病患者(对照)的混合血清孵育2 h,再加入二抗羊抗人IgG反应2 h,DAB显色。结果:分别建立了肺腺癌混合血清和对照混合血清孵育的肺腺癌组织双向蛋白印迹图谱,两个图谱之间存在明显差异。结论:利用混合血清孵育的肺腺癌组织总蛋白能建立良好的双向蛋白印迹图谱,这为进一步鉴定肺腺癌肿瘤相关抗原奠定了基础。
Aim:To establish the 2-dimensional Western blot maps of lung adenocarcinoma incubated with mixed se -ra.Methods:The soluble proteins of lung adenocarcinoma tissue samples from 2 patients were extracted and separated by 2-dimensional electrophoresis , then transferred onto nitrocellulous membranes .After having reacted with mixed sera from 4 lung adenocarcinoma patients or 4 patients with benign lung disease for 2 h, respectively , the nitrocellulous membranes were incubated with goat anti human IgG as the second antibody for 2 h, then DAB kit revealed the results of reaction .Re-sults:2-dimensional Western blot maps of lung adenocarcinoma incubated with mixed sera from lung adenocarcinoma and benign lung disease patients were established successfully , and the differences between the two maps were showed .Con-clusion:The excellent 2-dimensional Western blot maps of lung adenocarcinoma incubated with mixed sera can be built up, which will lay the foundation for the following identification of tumor-associated antigens in lung cancer .
