目的:采用反相高效液相色谱法测定不同厂家心安胶囊中绿原酸、牡荆素葡萄糖苷、牡荆素鼠李糖苷、牡荆素、芦丁和金丝桃苷的含量。方法:采用Agilent ZORBAX SB-C18(4.6mm×250mm,5μm)色谱柱;流动相为0.1%甲酸水溶液-乙腈-四氢呋喃,梯度洗脱;检测波长350nm,柱温30℃,流速1.0ml/min,进样量10μl。结果:绿原酸、牡荆素葡萄糖苷、牡荆素鼠李糖苷、牡荆素、芦丁和金丝桃苷的浓度在12.5-400、25-800、31.25-1000、9.38-300、2.50-80和9.38-300μg/ml范围内有良好的线性关系,该方法加样回收率平均为98.3%-100.3%。不同厂家心安胶囊6种指标成分的含量差异较大(RSD 11.0%-47.0%)。结论:该方法准确可靠、重现性好,可用于心安胶囊的质量控制。
Objective: To simultaneously determine the content of chlorogenic acid, itexin-glucoside, vitexin rhamnoside, vitexin, rutin and hyperoside in Xin'an Capsule from different manufacturers by RP-HPLC. Methods: Agilent ZORBAX SB-C18(4.6mm×250mm, 5μm) chromatographic column was used in this study: 0.1% formic acid-acetonitriletetrahydrofuran as mobile phase in gradient elution model, detection wave-350 nm, column temperature-30℃, fl ow rate-1.0ml/min, sample size-10μl. Results: The linear range of chlorogenic acid, itexin-glucoside, vitexin rhamnoside, vitexin, rutin and hyperoside was respectively 12.5-400, 25-800, 31.25-1000, 9.38-300, 2.50-80 and 9.38-300μg/ml. The average recovery rate was 98.3%-100.3%. The content of 6 components in Xin'an Capsule from different manufacturers was quite different(RSD 11.0%-47.0%). Conclusions: The method is simple and reproducible, and can be used for quality control of Xin'an capsules.
