目的:建立毛细管区带电泳内标法测定半夏鸟苷含量的方法。方法:采用毛细管区带电泳内标法定量,以熔融石英毛细管(57 cm×75μm,有效长度50 cm)为分离通道,运行缓冲液经NaOH溶液调pH至9.0的0.8 mol.L-1硼酸溶液-0.1mol.L-1硼砂溶液-乙腈(50∶50∶13),分离电压25 kV,检测波长254 nm,毛细管温度25℃,自动压力进样5 s。结果:半夏中鸟苷在0.25~1.5 mg线性关系良好(r=0.999 1),鸟苷加样回收率为102.0%,RSD为2.4%。结论:用毛细管区带电泳内标法测定半夏鸟苷含量的方法准确可行。
Objective:To develop the capillary zone electrophoresis(CZE) method for quantitative analysis of guanosine in Pinelliae Rhizoma using aciclovir as internal standard.Method:A fused silica capillary(57 cm×75 μm ID,an effective length of 50 cm) was used.The running buffer was composed of 0.8 mol·L-1 boric acid,0.1 mol·L-1 borate and acetonitrile(50:50:13) with pH adjusted to 9.0.The applied voltage was 25 kV and the capillary temperature was 25 ℃.The detection wavelength was at 254 nm.Result:Good linearity was found in the range of 0.25-1.5 mg for guanosine with r=0.999 1.The average recovery was 102.0% for guanosine with the relative standard deviation 2.4%(n=5).Conclusion:The method is simple and accurate to determine guanosine in Pinelliae Rhizoma.
