目的观察益气养阴活血药物人参、玉竹、川芎组合对体外培养脑缺血再灌注合并高糖培养条件下大鼠神经干细胞增殖的影响。方法神经干细胞培养分为4组:正常对照组、缺血再灌注模型组、缺血再灌注后高糖模型组、缺血再灌注后高糖加益气养阴活血药物血清组。BrdU掺入法观察神经干细胞的增殖,甲基噻唑基四唑比色试验检测细胞活性。结果脑缺血再灌注后神经干细胞增殖显著增加,细胞活力下降;高糖可抑制神经干细胞的增殖,降低细胞活力;益气养阴活血药物血清可促进神经干细胞的增殖,提高其活力。结论益气养阴活血药物可促进脑缺血再灌注后高糖培养大鼠神经干细胞的增殖,提高细胞活力。
Objective To study the effects of sera containing ginseng,fragrant solomonseal rhizome and ligustrazine,a prescription for supplementing qi,nourishing yin and activating blood circulation,on the proliferation of neural stem cells cultured in high concentration glucose medium from rats undergoing cerebral ischemia reperfusion.Methods The cultured neural stem cells were randomly divided into four groups: group A cultured in medium supplementing with normal rat sera,group B cultured in medium supplementing with sera from rats undergoing cerebral ischemia reperfusion,group C cultured in medium supplementing with high concentration glucose and sera from rats undergoing cerebral ischemia reperfusion,and group D cultured in medium supplementing with high concentration glucose and sera from rats undergoing cerebral ischemia reperfusion and treated with Chinese prescription for supplementing qi,nourishing yin and activating blood circulation.The proliferation of neural stem cells was observed using 5-bromodeoxyuridine incorporation.The activities of neural stem cells were observed using methyl thiazolyl tetrazolium colorimetry.Results The proliferation of neural stem cells from rats undergoing ischemia reperfusion increased significantly,and the cell activities significantly decreased.High concentration glucose could inhibit the proliferation of neural stem cells and reduced their activities.Sera containing Chinese prescription of supplementing qi,nourishing yin and activating blood circulation could promote the proliferation of neural stem cells and increase their activities.Conclusion Chinese prescription of supplementing qi,nourishing yin and activating blood circulation can promote the proliferation and the activities of neural stem cells cultured in high concentration glucose medium from rats undergoing cerebral ischemia reperfusion.
