目的:研究虫草素抑制脂多糖(LPS)诱导气道上皮细胞损伤作用机制。方法:培养气道上皮细胞9HTZ,50μg/ml的LPS干预9HTZ细胞8h后,采用四甲基偶氮唑蓝(MTT)法检测细胞活力,并收集细胞上清液和细胞,采用酶联免疫法(ELISA)法检测细胞上清液中的白介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β),采用Western blot法检测细胞中和转录因子NF-κB、细胞间粘附分子1(ICAM-1)蛋白表达。结果:虫草素(50,100μmol/ml)组能显著抑制LPS诱导的9HTZ细胞的凋亡,并能显著性抑制细胞上清液中的IL-6、IL-1β、TNF-α的含量,抑制细胞中NF-κB、ICAM-1蛋白的表达。讨论:虫草素能保护LPS诱导的气道上皮细胞损伤。
To investigate the effect of eordycepin on LPS-indueed injury in 9HTZ cells and its mechanism. Methods: Cultured airway epithelial cells 9HTZ, LPS (50μg/ml) on 9HTZ cells for 8h, using four methyl thiazolyl tetrazolium (MTT) assay for cell viability, and cell supematant and eels were collected, using enzyme linked immunosorbent assay (ELISA) method was used to detect cell supematants of in- tedeukin-6 (IL-6), tumor necrosis factor alpha (TNF- α), interlenkin-1 beta (IL-1μ), the transcription factor NF- κ B, intercellular adhe- sion molecule 1 (ICAM-1) protein expression were detected by Western blot. Results: Cordyeepin (50,100μmoi/ml) inhibited cell apopto- sis in LPS-induced 9HTZ cells ( P 〈 0.01 ), and can significantly inhibit the cell content of IL-6, IL-1μ, TNF-α ( P 〈 0.01 ) in the supema- rant, inhibiedt the expression of NF-κB, ICAM-1 proteins in cells (P 〈 0.01 ). Conclusion: The eordycepin can protect injury in LPS-in- duced cells.
