目的建立同时测定广东紫珠中熊果酸和齐墩果酸含有量的HPLC法,并对不同产地、采收期、部位中的这两种成分进行比较。方法采用ZOBAX Eclipse XDB-C18色谱柱(4.6 mm×250 mm,5μm);甲醇-0.1%磷酸水溶液(90∶10)为流动相;体积流量1 m L/min;检测波长210 nm;柱温25℃。结果熊果酸、齐墩果酸线性范围分别为0.052 4~1.048μg(r=0.999 8)、0.055~1.107μg(r=0.999 8),平均加样回收率分别为99.9%、100.7%,RSD分别为0.8%、1.1%。结论广东紫珠中熊果酸和齐墩果酸的含有量以江西萍乡芦溪产者最高。随着生长期延长,两者含有量呈先升高后降低的趋势,并在9月份最高。而且,在不同部位中,其分布规律均为果实〉叶〉茎〉根。
AIM To establish a method for simultaneously determining the contents of ursolic acid and oleanolic acid in Callicarpa kwangtungensis Chun. by HPLC and compare these two constituents in different growing areas,picking times and parts. METHODS The analysis was performed on ZOBAX Eclipse XDB-C18column( 4. 6mm × 250 mm,5 μm),mobile phase was methanol-0. 1% phosphoric acid aqueous solution( 90 ∶ 10),flow rate was 1. 0 m L / min,detection wavelength was set at 210 nm,and column temperature was maintained at 25 ℃. RESULTS The linear ranges of ursolic acid and oleanolic acid were within 0. 052 4- 1. 048 μg( r = 0. 999 8) and0. 055- 1. 107 μg( r = 0. 999 8),and the average recoveries( n = 5) were 99. 9% and 100. 7% with the RSDs of0. 8% and 1. 1%,respectively. CONCLUSION The C. kwangtungensis sample from Luxi( Pingxiang City,Jiangxi Province) has the highest contents of ursolic acid and oleanolic acid. As growth time prolongs,the contents of these two constituents increase at first and then decrease. In addition,the distribution law of them is fruit〉leaf〉stem〉 root.
