目的通过观察EphrinB2对小鼠妊娠中期子宫血管内皮生长因子(VEGF)表达及孕体生长情况的影响来探讨EphrinB2促血管发育的机制。方法C57BL/6小鼠分为4组,分别为1组(对照组)、2组(低浓度组,EphrinB2-Fc10μg/kg,ip)、3组(中浓度组,EphrinB2-Fc50μg/kg,ip)和4组(高浓度组,EphrinB2-Fc250μg/kg浓度组,ip),2、3、4组在检查到阴栓当天腹腔注射EphrinB2-FcPBS溶液,对照组注射等量PBS,连续注射7d,妊娠10.5d处死小鼠,称量孕体重量,应用实时聚合酶链反应(RealtimePCR)及免疫组化法检测子宫VEGFmRNA和蛋白的表达。结果(1)小鼠孕体重量在Eph—rinB2高浓度组明显增高;(2)EphrinB2促进小鼠子宫VEGFmRNA及蛋白表达。结论EphrinB2调控妊娠血管发育的作用可能与其促子宫VEGF表达增加有关,EphrinB2亦可促进孕中期孕体生长加速。
Objective: To investigate the effect mechanism of EphrinB2 on the vascular development by observing the effect of EphrinB2 on the VEGF expression of the uterus and the coneeptus growth in middle of the gestation of mice. Methods: Twelve pregnant C57BL/6 mice were randomly divided into four groups. The mice in group 1 (control group) were treated with phosphate buffered saline (PBS). The mice in group 2, group 3 and group 4 were treated with 10 gg/kg, 50 9g/kg and 250 9g/kg of EphrinB2-Fc by intraperitoneal injection respectively. The day that the plug was seen was 0. 5 day after gestation. The pregnant mice were injected intraperitoneally EphrinB2-Fc from 0. 5 d for 7 days and sacrificed at 10. 5 d after gestation. The conceptuses were weighed. Real-time PCR and immunohistochemistry assay were applied to examine the expression of mRNA and protein of VEGF in the uterus. Results:The weights of the conceptuses in group 3 and group 4 were higher than those of control group. The expression of VEGF was significantly promoted by EphrinB2 (P〈0.05). Conclusions: EprhrinB2 regulated the vascular development by increasing the expression of VEGF. Moreover, EprhrinB2 promoted the growth of the conceptus. Further study of the mechanism of EphinB2 on the pregnancy was needed.
