目的探讨慢性焦虑刺激对成年雌性大鼠性激素水平的影响。方法将雌性成年大鼠随机分成空白组、模型组,采用不确定性空瓶刺激法建立焦虑应激大鼠模型。以高架十字迷宫实验评价其行为学;采用化学发光法检测大鼠性激素六项:促黄体生成素(LH),促卵泡激素(FSH),泌乳素(PRL),睾酮(T),孕酮(P),雌二醇(E2)水平;光镜观察大鼠子宫、卵巢的形态学变化。结果与空白组相比,模型组大鼠血清T浓度显著升高(P〈0.01),LH浓度显著减少(P〈0.01),FSH、PRL、P、E2水平差异无统计学意义;模型组卵巢组织闭锁卵泡多,黄体和成熟卵泡少,子宫内膜较薄,间质纤维增生。结论在焦虑状态下,成年雌性大鼠的血清T浓度升高,LH浓度降低,子宫、卵巢出现病理改变,提示焦虑是影响雌性大鼠生殖机能的重要因素。
Objective To study the effects of chronic anxiety stimulation on sex hormones levels in the female adult rats. Methods 20 female adult rats were randomly allocated into model group and blank group. The anxiety stress model was established by uncertainty empty bottle stimulation. The behavioral changes of the rats were measured by evaluated plus-maze test. The sex hormones (LH, FSH, PRL, T, P and E2) of female rats were determined by chemiluminescence method. The uterus and ovaries histomorphological changes were observed by light microscope. Results Compared with blank group, the serum T of model group increased obviously (P〈0.01), while the LH decrease lower LH (P〈O.01), and the levels of FSH, PRL, P, and E2 were not statistically significant. More atretic follicle, less corpus luteum and mature follicle, thinner endometrium and stromal fibrous proliferation were showed in the model group. Conclusion In the anxiety state, the serum T concentration in female adult rats was significantly increased, while the LH level decreased Some pathologic changes occurred in uterus and ovary of the rats. So anxiety is an important factor affecting the reproductive function.
