无色花色素还原酶(leucoanthocyantin reducase,LAR)是植物类黄酮生物合成途径中的一个关键酶。本研究根据海南龙血树(Dracaena cambodiana)转录组数据,利用RT-PCR技术克隆1个编码无色花色素还原酶基因DcLAR1,结果表明:该基因全长1 355 bp,包含一个1 011bp的开放阅读框,编码336个氨基酸。DcLAR1蛋白具有典型的植物无色花色素还原酶结构特征,包含3个保守的结构域RFLP、ICCN和THD。表达分析结果表明,DcLAR1在根、茎、花、叶和果中具有不同程度的表达;DcLAR1表达受到血竭诱导剂的诱导,与血竭积累正相关,推测该基因可能在海南龙血树类黄酮生物合成过程中具有重要功能。
The leucoanthocyantin reducase (LAR) is a key enzyme in plant flavonoid biosynthesis pathway. In this study, we based on transcriptome data ofDracaena cambodiana to clone 1 coded leucoanthocyanidin reductase gene named DcLA R1 by using RT-PCR technology. The result showed that the full-length of cDNA was 1 355 bp, cont- aining 1 011 bp opening reading flame (ORF), encoding 336 amino acids. DcLAR1 protein had the typical structural feature of plant leucoanthocyantin reducase, containing three conservative structure domain RFLP, ICCN and THD. The expression results showed that DcLAR! was differently expressed in root, stem, fower, leaf and fruit; the expression of DcLAR1 was induced by dragon's blood inducers, positively related to the accumulation of dragon's blood. We speculated that the DcLAR1 may play an important role in flavonoid biosynthesis process in Dracaena cambodiana.
