目的探讨JWA基因在不同转移潜能人肝癌细胞中的表达及意义。方法分别应用荧光定量RT-PCR和Western blot法检测无转移潜能人肝癌细胞株Hep G2、低转移潜能人肝癌细胞株MHCC97L、高转移潜能人肝癌细胞株MHCC97H及较高转移潜能人肝癌细胞株HCCLM3的JWA mRNA和蛋白表达水平。结果 Hep G2细胞JWA mRNA和蛋白水平最高,HCCLM3细胞的JWA mRNA和蛋白水平最低,JWA基因表达水平随着肝癌细胞转移潜能增高而递减。结论 JWA mRNA和蛋白表达水平随着肝癌细胞转移潜能增高而递减,可能与肝癌的发生、发展及转移密切相关。
Objective:To explore the expression and significance of JWA gene in human hepatocellular carcinoma(HCC) cells with different metastatic potentials.Methods:The expression levels of JWA protein and its mRNA were detected in stepwise metastatic HCC cell lines of Hep G2,MHCC97L and MHCC97H by quantitative real-time PCR and Western blot.Results:The highest expression levels of JWA protein and its mRNA in Hep G2 cells were seen and the lowest expression levels of JWA protein and its mRNA in HCCLM3 cells were found.The expression levels of JWA gene were decreased as metastatic potential increased.Conclusion:The expression levels of JWA protein and its mRNA are inversely correlated with the metastatic potential of HCC cells and may be closely correlated to the oncogenesis,development and metastasis of HCC.
