甜菜M14品系具有无融合生殖特性,为了获得M14品系特异表达的基因,本实验室前期利用抑制消减杂交(SSH)和mRNA差异显示(DDRT-PCR)技术对M14品系和能够进行有性生殖的栽培甜菜A2Y品系进行了基因特异表达研究,获得了298条M14品系特异表达基因的ES-Ts。对这些ESTs进行生物信息学分析并进行了GO分类,筛选出5条可能与基因表达调控和细胞周期调控相关的ESTs,为进一步获得这些基因的全长序列并研究其功能奠定了基础,也为今后能在甜菜M14品系中寻找并克隆与无融合生殖相关的基因提供了可靠的保证。
M14 is a monosomic addition line with the characteristic of apomixis. By using the methods of suppression subtractive hybridization ( SSH ) and mRNA differential display reverse transcriptional PCR ( DDRT - PCR) between M14 and A2Y, 298 ESTs of specially expressed genes of M14 were obtained and the relevant bioinformatics analysis was made. Then, they were annotated and classified by GO ( Gene ontology) method, 5 ESTs whose function were closely related to regulation of gene expression and cell cycle. The research is the basis for obtaining full - length cDNA of these ESTs and characterizing their protein functions in the further study.
