目的探讨金银花提取物对化合物48/80、组胺及P物质诱导的小鼠瘙痒模型的作用。方法采用0.3、0.8、2.4 g/kg 3个不同剂量的金银花提取物将小鼠灌胃给药1 h后,分别皮内注射致痒物质化合物48/80、组胺和P物质,随后将小鼠置于观察笼内,用摄像仪记录1 h内小鼠的行为。结果金银花提取物可呈剂量依赖性的抑制P物质诱导的小鼠瘙痒,而对化合物48/80和组胺诱导的小鼠瘙痒没有明显的抑制作用。结论金银花提取物对P物质诱导的小鼠模型有明显的抗瘙痒作用,其抗瘙痒活性主要作用部位为角质形成细胞,而非肥大细胞。
Objective To study the effect of Flos Lonicerae Japonicae on pruritus induced by various pruritic agents including compound 48 /80,histamine and substance P in mice. Methods Flos Lonicerae Japonicae extracts( 0. 3 g / kg,0. 8 g / kg and 2. 4 g / kg) were respectively administered orally to ICR mice 1 h before the injection of compound 48 /80,histamine and substance P. Immediately after the injection of pruritogens,the animals were put into the acrylic cage again,and their behaviors were videotaped. Results Flos Lonicerae Japonicae dose-dependently inhibited substance P-induced scratching. However,no inhibitory effect was observed on the scratching behavior induced by histamine or compound 48 /80. Conclusion Flos Lonicerae Japonicae shows significant antipruritic effect on substance P-induced pruritus in ICR mice. The primary site of its antipruritic activity is epidermal keratinocytes rather than mast cells.
