目的:探究黄精多糖(Polygonatum sibiricum polysaccharides,PSP)对H9c2心肌细胞缺氧/复氧(hypoxia-reoxygenation,H/R)损伤的保护作用。方法:体外无菌培养H9c2心肌细胞,制备缺氧/复氧损伤模型。应用A0-EB和Hoechst33324染色法观察心肌细胞凋亡形态学变化,流式细胞仪检测细胞凋亡率,采用MTT法检测心肌细胞存活率,Westen Blot法检测H9c2心肌细胞Caspase-3、Bcl-2及Bax蛋白表达。结果:缺氧/复氧模型组中H9c2细胞存活率明显降低,细胞核浓染致密、固缩,细胞凋亡率明显增加,Caspase-3的表达和Bax/Bcl-2比率显著上调;与模型组比较,黄精多糖1.0、1.5、2.0mg/ml药物组中细胞存活率均明显升高,细胞核淡染,轻微缩小,细胞凋亡率降低,Bax/Bcl-2比率明显下调,Caspase-3表达水平被显著抑制。结论:黄精多糖对H/R诱导的H9c2心肌细胞具有保护作用,其作用可能与抑制心肌细胞凋亡有关。
Objective: To study the effects of Polygonatum sibiricum polysaccharides( PSP) on H9c2 myocardial cells injury induced by hypoxia/reoxygenation.Methods: H9c2 cells were treated with hypoxia-reoxygenation to establish a myocardial injury in vitro model.Cell viability was measured by methylthiazolyl tetrazolium( MTT),Hoechst33324 and AO-EB fluorescent staining.Annexin V-FITC flow cytometry detected apoptosis,Western blot was performed to detect the expression of Caspase-3,Bax and Bcl-2 protein levels.Results: The survival rate of myocardial H9c2 cells in H/R group significantly reduced.Chromatin cell nucleus was dyed densely and compact,presenting blue-white.Compared with H/R group,the survival rates of myocardial H9c2 cells in each PSP group significantly increased.And the apoptosis rates and expression of Caspase-3,Bax,Bax/Bcl-2 were remarkably down-regulated,but Bcl-2 was remarkably up-regulated.Conclusion: PSP could protect H9c2 cells against H/R impairment,and its protection is possibly related to the reduction apoptosis.
