目的:探讨干细胞生长的微环境。 资料来源:应用计算机检索Pubmed1975-01/2004-12和Ovid 1975-01/2004-12以及Elserve 1992-01/2004-12数据库中与干细胞微环境相关的文章,检索词为“neural stem cells,niche or microenvironment,or transplantation”,并限定文章语言种类为English。同时检索维普中文科技期刊数据库1990-01/2004-12相关干细胞微环境的文章,检索词为“神经干细胞、微环境或移植”,并限定文章语言种类为中文。 资料选择:对资料进行初审,开始查找全文,纳入标准:①近5年文献。②有关干细胞、移植与其微环境关系的文献。排除标准:重复研究、Meta分析和综述类文献。 资料提炼:收集了近50篇文章,纳入22篇符合标准的文章进行综述。资料综合:干细胞的自我更新、增殖和分化主要依赖于周围特殊的微环境,即小生境。干细胞小生境可能提供了一种调节体内干细胞数量的手段。小生境不是几种细胞简单的集合体,而是一个有机的组织结构、生理功能整体。成人神经发生是在血管原性的小生境中进行的。基底膜在干细胞小生境中起固定各种因子、细胞和提供各种空间信号的作用。 结论:小生境与神经干细胞的生存、分裂、分化及迁移均有关系,要求在移植时尽量满足干细胞的生长微环境。
OBJECTIVE: To discuss the niche or microenvironment of neural stem cells. DATA SOURCES: A computer-based online search of Pubmed and Ovid databases between January 1975 and December 2004 and Elserve database between January 1992 and December 2004 was undertaken to identify English literatures about niche of neural stem cells by using the key words of "niche or microenvironment, neural stem cells, or transplantation". Meanwhile, the Chinese literatures in the Vip Chinese Periodical database published from January 1998 to December 2004 were searched with the same key words. STUDY SELECTION: The articles were selected firstly and searched the full text. Inclusive criteria: ①Articles published in recent 5 years. ②Articles relevant to neural stem cells, transplantation and its niche or microenvironment. Exclusive criteria: repetitive research, Meta analysis and review articles. DATA EXTRACTION: A total of 50 papers were collected, and 22 ones accorded with the criteria were reviewed. DATA SYNTHESIS: The self-renewal, proliferation and differentiation of neural stem cells mainly depend on surrounding specific microenvironment, named niche, which may provide a method for the regulation of the number of stem cells. Niche is not the simple aggregen of several kinds of cells, but an organic whole of organization, physiological function. Adult neurogeny occurs in the niche of blood vessel. The basal lamina can fix various cytokine, cells and provide various spatial signals in niche of stem cells. CONCLUSION: Niche is correlated with survival,division,differentiation and migration of neural stem cells, therefore it is necessary to satisfy the niche of neural stem cells when transplantation.
