目的:研究中药复方肺岩宁方对上皮-间质细胞转化(epithelial—mesenchymal transition,EMT)因子Twist、Snail、Sip1及E—cadherin mRNA表达的影响。方法:选用C57小鼠Lewis肺癌转移肿瘤模型,采用实时PCR的方法观察肺岩宁方作用前后肿瘤组织和转移肺组织中Twist、Snail、Sip1及E—cadherin的mRNA表达。结果:Twist、Snail在C57 Lewis肺癌转移肺组织中mRNA表达较正常C57小鼠肺组织中明显增高(P〈0.01),Sip1则无明显差异(P〉0.05),而E—cadherin mRNA表达降低(P〈0.01);C57 Lewis肺癌小鼠经过肺岩宁方治疗后,Twist、Snail在肿瘤组织和转移肺组织中mRNA表达较模型组明显降低(P〈0.05)。而E—cadherin的表达增高(P〈0.01),但Sip1 mRNA表达在肺岩宁方治疗前后无明显变化(P〉0.05)。结论:在C57小鼠Lewis肺癌转移肿瘤模型中证实了上皮-间质细胞转化是肿瘤转移的关键步骤之一,调节上皮-间质细胞转化核转录因子Twist、Snail和标志因子E—cadherin是肺岩宁方抗肿瘤转移的重要途径之一。
Objective : To study the effect of traditional Chinese medicine Feiyanning Decoction on mRNA expressions of eptithelial-mesenchymal transition (EMT)-related factors such as Twist, Snail, Sip 1, and E-cadherin. Methods:Xenograft model of Lewis metastatic lung carcinoma was established in C57 mice. The effects of Feiyanning Decoction on mRNA expression of Twist, Snail, Sip 1, and E-cadherin in tumor tissues and metastatic lung tissues were detected by real-time PCR. Results: The mRNA expressions of Twist and Snail in metastatic lung tissues were significantly higher than that in the control group ( P 〈0.01 ). There was no significant difference in the expression of Sip 1 between the two groups ( P 〉 0.05 ), while the expression of E-cadherin was significantly decreased (P 〈0.01 ). After treatment with Feiyanning Decoction, the mRNA expressions of Twist and Snail in tumor tissues and metastatic lung tissues were significantly decreased compared with the control group (P 〈0.05 ) , while the expression of E-cadherin was increased significantly (P 〈 0.01 ), but there was no significant difference in the expression of Sip 1 between two groups (P 〉 0.05 ). Conclusion: The fact that EMT is the key step of tumor metastasis was confirmed in C57 mice with Lewis metastatic lung carcinoma. Regulation of EMT- related factors Twist, Snail, and marker E-cadherin is one of the important pathways for anti-metastatic effects of Feiyaning Decoction.
