目的:研究南方红豆杉水提物联合紫杉醇对肺癌A549细胞增殖的抑制作用和机制。方法:将A549细胞分为低浓度紫杉醇联合不同浓度红豆杉组、低浓度紫杉醇组、空白对照组,用CCK-8法检测药物作用48h后对细胞存活率的影响,用Western blot检测细胞内Bcl-2、Bax、Survivin蛋白表达变化。结果:红豆杉可增加紫杉醇对A549细胞的抑制作用,且随着药物浓度的增加,其抑制作用逐步增强;紫杉醇联用红豆杉可通过下调Bcl-2,Survivin,并上调Bax的表达,而促进细胞发生凋亡。结论:红豆杉联合紫杉醇能抑制A549细胞生长,其机制可能与凋亡相关基因Bcl-2,Survivin的降低,促凋亡基因Bax的增高有关。
Objective: To study the inhibitory effect of aqueous extract of Taxus chinensis var. mairei combining with paclitaxel on human lung carcinoma A549 cells, and to explore its mechanism. Methods: According to the results of the pre experiment, the A549 cells were divided into different concentrations of paclitaxel groups, different concentrations of Taxus chinensisvar. mairei groups and blank group. The drug effect of 48 h and the effect on cell survival were detected by the method of CCK-8. Based on the above results, then A549 cells were divided into low concentration of paclitaxel combining different concentrations of Taxus chinensis var. mairei groups, low concentration of paclitaxel group, blank control group, the drug effect of 48 h and the effect on cells survival were detected by the method of CCK-8. The protein expression of Bcl-2, Bax and Survivin were detected by Western blot. Results: Taxus chinensis var. mairei combining paclitaxel inhibited the proliferation of A549 cells, and with the increase of drug concentration, the inhibitory effect gradually increased; Aqueous extract of Taxus chinensis var. mairei combining with paclitaxel promoted cells apoptosis by down-regulating the expression of Bcl-2, Survivin and up-regulating the expression of Bax. Conclusion: The mechanism underlying Taxus chinensis var. mairei combining paclitaxel inhibiting the growth of A549 cells may be correlative with an decrease of Bcl-2 and Survivin related to apoptosis genes and an increase of apoptosis gene Bax.
