目的观察抑制CLC-2基因的表达对人眼小梁细胞细胞周期进程的影响。方法构建针对CLC-2的小干扰(siRNA)重组表达载体,脂质体LipofectamineTM2000介导转染人眼小梁细胞后,应用RT-PCR半定量检测小梁细胞CLC-2mRNA表达量的变化;流式细胞仪检测干扰CLC-2表达后,人眼小梁细胞细胞周期进程。结果与空载体组比较,重组载体组的CLC-2mRNA表达明显降低,且有效抑制小梁细胞由G0期到G1的转位。结论抑制CLC-2的表达可以干扰人眼小梁细胞正常细胞周期进程。
Objective To investigate the effect of CLC-2 suppression by antisense oligonucleotide on cell cycle of human trabecular meshwork cells.Methods CLC-2 mRNA-targeted hairpin siRNA was designed and constructed,and transfected into cultured human trabecular meshwork cells by LipofectaminTM2000.RT-PCR semi-quantitative was applied to detect the expression of the cell′s CLC-2 gene,and flow cytometry was performed to detect the change of cell cycle on interfered human trabecular meshwork cells.Results The expression of CLC-2 gene in siRNA interfered groups was significantly lower than that of control group,and the cell cycle of siRNA interfered human trabecular meshwork cells was effectively inhibited from GO to G1 phase.Conclusions To inhibit the expression of CLC-2 can effectively interfere cell cycle of the human trabecular meshwork cells.
