目的探讨CXCR4/SDF-1α信号在乳腺癌细胞体外增殖和迁移中的作用。方法免疫组织化学染色法检测CXCR4在乳腺癌组织中的表达;采用免疫荧光标记法和RT-PCR方法检测CXCR4在乳腺癌细胞株上的表达;MTT法研究SDF-1α细胞因子对乳腺癌细胞株体外增殖的影响及抗体12G5的阻断作用;体外微孔隔离室迁移技术研究CXCR4/SDF-1α信号对乳腺癌细胞株体外迁移能力的影响和抗体12G5的阻断作用。结果 CXCR4表达于乳腺癌组织,也在乳腺癌细胞株表达。SDF-1α细胞因子可有效促进乳腺癌细胞株MDA-M231的体外增殖和迁移,而阻断型CXCR4单抗12G5可有效抑制其增殖和迁移,并呈浓度依赖性。结论 CXCR4/SDF-1α信号参与了乳腺癌细胞株的体外生长和转移,与乳腺癌的侵袭和转移有关。
Objective To investigate the role of CXCR4/SDF-1α signal in the proliferation and migration of breast cancer cell lines in vitro.Methods Immunohistochemistry method was used to detect the CXCR4 expression in breast cancer tissue.Immunofluorescence staining and RT-PCR methods were used to detect the CXCR4 expression on the breast cancer cell lines.The proliferation of breast cancer cell line MDA-M231 and the blocking effect of 12G5 on the proliferation were analyzed by MTT method.The role of CXCR4/SDF-1α in the MDA-M231 cells migration and the blocking effect of 12G5 antibodies were analyzed by transwell assay.Results CXCR4 expression was found in breast cancer tissue,as well as on the breast cancer cell lines.SDF-1α could stimulate the proliferation and migration of MDA-M231 cells in vitro.However,CXCR4 specific functional monoclonal antibody 12G5 could block that proliferation and migration stimulated by SDF-1α with a dose-dependent manner.Conclusions CXCR4/SDF-1α signal is involved in the growth and migration of breast cancer cells in vitro.It suggests that the signal is related to the metastasis and invasion of breast cancer in vivo.
