目的探讨曲格列酮对大鼠系膜细胞(mesangial cell,MC)增殖和凋亡的影响。方法以体外培养大鼠系膜细胞为研究对象,应用不同浓度曲格列酮刺激系膜细胞。采用流式细胞仪检测系膜细胞凋亡,并用DNA倍体分析检测细胞周期,采用锥虫蓝染色检测系膜细胞增殖。结果系膜细胞经低浓度曲格列酮(1、5μmol/L)干预后出现明显凋亡,与正常对照组相比,差异有统计学意义(均P〈0.05)。经不同浓度曲格列酮干预后,系膜细胞增殖明显抑制,与正常对照组相比,差异有统计学意义(均P〈0.05)。同时5μmol/L曲格列酮干预系膜细胞后可将细胞周期阻滞于G2/M期。结论曲格列酮可明显抑制系膜细胞增殖,促进系膜细胞凋亡。
Objective To investigate the effects of troglitazone on the apoptosis and proliferation of rat mesangial cells. Methods Rat mesangial cells were incubated with different concentrations of troglitazone. The apoptosis of mesangial cells was measured by flow cytometry. Measurement of cellular DNA content and the analysis of the cell cycle were performed by flow cytometry. Cell proliferation was assessed by MTT colorimetric assay. Results The apoptosis of mesangial cells was increased after exposure to 1 and 5 μmol/L troglitazone as compared with normal control group (P〈0.05). The proliferation of mesangial cells was also inhibited by stimulation with 1 and 5μmol/L troglitazone (P〈0.05). 5 μmol/L troglitazone made the cell cycle arrest in G2/M phase. Conclusion Troglitazone could significantly inhibit the proliferation of mesangial cells and make mesangial ceils arrest in G2/M phase and induce the cell apoptosis.
